Abstract
In this study, the potential of anti-cancer compounds production against lung cancer cells was investigated in actinomycete strains which were taken from University of Tehran’s Microorganisms Collection (UTMC).
The number of 40 actinomycete strains which were stored under -70 oC, were selected randomly. After sporulation period, seeding and stages fermentation of each strain were prepared. Extracted metabolites of each strain were deposited at University of Tehran’s Biocompound Collection (UTBC) and then Toxicity level of crude extracts was measured by Brine Shrimp assay and MTT. The results showed that products of strains UTMC 863, UTMC 877, UTMC 919, UTMC 638 and UTMC 676 had mortality effects on A549 cell line. Anti-cancer producing strains were identified by morphological and molecular methods. 16S rRNA sequence analysis showed that strain UTMC 676 has 99.86% similarity to Streptomyces aureoverticillatus (Accession Number AY999774) and strain UTMC 638 has 98.78% similarity to Streptomyces cacaoi subsp. Cacaoi (Accession Number AB184115), also strains UTMC 863 and UTMC 877 have 99.85% and 100% similarity to Nocardia carnea (Accession Number BAFV01000017) respectively, likewise strain UTMC 919 has 99.29% similarity to Kribbella sancticallisti (AM778577). Considering the capable of the actinomycete in production of anti-cancer compounds, extracts of strain with this capability obtain in this project showed a good anti-cancer activity the need to discover new drugs due to drug resistant cancers, purification of these metabolites is proposed,
